Actions Microbiology Amoxycillin is a semisynthetic, bactericidal beta-lactam antibiotic in the amino-penicillin family that is used to treat susceptible, multiplying, Gram-positive and Gram-negative bacteria, including Actinobacillus sp., Actinomyces bovis, Bacillus anthracis, Bordetella sp., Clostridium sp., Corynebacterium sp., Enterococcus faecalis, Erysipelothrix rhusiopathiae, Escherichia coli, Fusiformis sp., Fusobacterium necrophorum, Haemophilus sp., Moraxella sp., Pasteurella sp., Proteus mirabilis, Salmonella sp., Streptococcus sp., Staphylococcus sp. and obligate anaerobes. Amoxycillin is not resistant to beta-lactamases produced by resistant bacteria and will therefore not be active on these strains.Pharmacology Amoxycillin impairs the development of bacterial cell walls. At concentrations lower than the minimum inhibitory concentration, the beta-lactam antibiotics exert a residual effect on bacterial structure and function which in turn promotes phagocytosis. These antibiotics are only effective against multiplying organisms. Amoxycillin is stable in gastric acid and may be given without regard to meals. Absorption is greater than 60%, and amoxycillin is widely distributed into most body fluids, urine, bile, lung and bone, but not intracellular spaces, the central nervous system or the eye. Use of amoxycillin in pregnancy is considered as Category B: low risk, and is excreted into milk at low levels.Pharmacokinetics Amoxycillin is inactivated by the liver to inactive penicilloic acids, and 40 to 70% is excreted unchanged in urine. The half-life is 45 to 90 minutes in dogs and 1 to 1.2 hours in cats, but this is extended with renal failure. Excretion occurs primarily via the urinary and biliary systems. Urinary excretion is via tubular secretion and glomerular filtration.
Precautions Use with caution in animals with renal failure. Consider reducing dose frequency to twice or once per day. Use with caution in lactating animals as amoxycillin can be excreted into milk.First Aid If poisoning occurs, contact a doctor or Poisons Information Centre. Phone Australia 131 126.
Dosage and Administration Dogs, cats. 4 to 11 mg/kg bodyweight orally twice daily until 48 hours after clinical signs have subsided.Amoxycillin is stable in gastric acid and may be given without regard to meals.
