Indication | Bacterial infections of the respiratory, musculoskeletal and urinary systems, soft tissue, mouth and skin when organisms sensitive to the combination of amoxycillin and clavulanic acid are suspected. |
Contraindications | Administration to small herbivores such as rabbits and guinea pigs, or to animals that have a history of allergic reactions to penicillins. |
Actions | Amoxyclav 250 is a broad spectrum bactericidal antibiotic tablet for dogs and cats with activity against many Gram-positive and Gram-negative organisms.Amoxycillin-clavulanic acid combination products have demonstrated bactericidal activity against Actinobacillus sp., Actinomyces bovis, Bacillus anthracis, Bordetella sp., Clostridium sp., Corynebacterium sp., Enterococcus faecalis, Erysipelothrix rhusiopathiae, Escherichia coli, Fusiformis sp., Fusobacterium necrophorum, Haemophilus sp., Moraxella sp., Pasteurella sp., Proteus mirabilis, Salmonella sp., Streptococcus sp., Staphylococcus sp. and obligate anaerobes.Microbiology The combination of amoxycillin and clavulanic acid overcomes beta-lactamase resistance issues that can occur with uncombined penicillins. Amoxycillin inhibits biosynthesis of cell wall mucopeptide in sensitive microorganisms but is not resistant to beta-lactamases and therefore has no activity against beta-lactamase producing organisms on its own. Potassium clavulanate is a potent progressive beta-lactamase enzyme inhibitor, inhibiting beta-lactams produced by Gram-positive and Gram-negative organisms. Potassium clavulanate is able to permeate into cell walls and so can inactivate extracellular enzymes and those bound to cell walls. On its own, potassium clavulanate has weak bactericidal activity but it provides synergistic antibacterial activity when administered in combination with amoxycillin trihydrate.Pharmacology Amoxyclav is well absorbed from the gastrointestinal tract after oral administration and can be given without regard to meals/feeding. Absorption is greater than 60%, and the amoxycillin moiety is widely distributed into most body fluids, urine, bile, lung and bone, but not intracellular spaces, the central nervous system or the eye.Pharmacokinetics Amoxycillin is inactivated by the liver to inactive penicilloic acids and 40 to 70% is excreted unchanged in urine. Clavulanic acid is metabolised by the liver and excreted via the urine, faeces and small amounts in expired air. The half-life is around 1 hour in dogs, and is extended with renal failure. There is evidence that giving amoxycillin with clavulanate has little effect on the pharmacokinetics of either medication. Use of Amoxyclav in pregnancy is considered as Category B: low risk, and is excreted into milk at low levels. |

Dosage and Administration | 12.5 mg/kg bodyweight twice daily. Dose rates of up to 25 mg/kg bodyweight twice daily have been shown to be effective in refractory cases. Duration of therapy is typically five to seven days, or for 48 hours after clinical signs have subsided. Longer duration of therapy is recommended for chronic cystitis and dermatological conditions.The combination is stable in gastric acid, and fate and absorption are not influenced by gastric or intestinal contents. This means it can be administered without regard to meals, and is appropriate for inappetent animals. |
