|Drug Name and Strength||Pimobendan 2.5mg|
|Indication||Treatment of canine congestive heart failure originating from valvular insufficiency (mitral and/or tricuspid regurgitation) or dilated cardiomyopathy.|
|Contraindications||This product is contraindicated in cases of hypertrophic cardiomyopathies or clinical conditions where an augmentation of cardiac output is not recommended for functional or anatomical reasons (e.g. aortic stenosis).|
|Actions||Pharmacology Pimobendan, a benzimidazole-pyridazinone derivative, is a nonsympathomimetic, nonglycoside inotropic substance with potent vasodilatory properties. It exerts its stimulatory myocardial effect by a dual mechanism of action: increase in calcium sensitivity of cardiac myofilaments and inhibition of phosphodiesterase (type III). It also exhibits a vasodilatory action through an inhibitory action on phosphodiesterase III activity.Pharmacokinetics After oral administration of pimobendan, the absolute bioavailability of the active principle is 60 to 63%. The mean plasma protein binding is 93%. The plasma elimination half-life of pimobendan is approximately 30 minutes and the main active metabolite elimination half-life is approximately two hours. Almost the entire dose is eliminated via faeces. Pimobendan absorption is affected by food. The stomach should be empty at the time of dosing. Do not administer with food and do not feed for at least one hour after dosing. In rat experiments it has been shown that pimobendan is excreted into milk.|
|Precautions||Use during pregnancy and lactation. Cardisure Flavoured Tablets should only be administered to pregnant and lactating bitches if the expected therapeutic benefits outweigh the potential risk. In studies with rats and rabbits, pimobendan had no effect on fertility and embryotoxic effects only occurred at maternotoxic doses.Studies into the effect of pimobendan on the reproductive function of male dogs have not been conducted. Administration to dogs not showing signs of cardiac disease is not recommended.First Aid If poisoning occurs, contact a doctor or Poisons Information Centre. Phone Australia 131 126. Accidental ingestion, especially by a child, may lead to the occurrence of tachycardia, orthostatic hypotension, flushing of the face and headaches.|
|Dosage and Administration||Administer tablets orally at a dose range of pimobendan 0.2 mg to 0.6 mg/kg bodyweight per day. The preferable daily dose is pimobendan 0.5 mg/kg bodyweight. The dose should be divided into two administrations (0.25 mg/kg bodyweight), one half dose in the morning and the other half approximately 12 hours later.The maintenance dose should be individually adjusted according to the severity of the disease. Each dose should be administered on an empty stomach at least one hour before feeding.Tablets are deeply scored; to break a tablet into two halves, place the tablet on an even surface with the scored side up, hold one half of the tablet and press down on the other half. To break a double scored tablet into quarters, place the tablet on an even surface with the scored side up and apply pressure on the middle with your thumb.Monitoring of cardiac function and morphology is recommended in animals treated with pimobendan.|
|Storage||Store below 25°C (air conditioning). Protect from light. Do not expose to elevated temperatures above 30°C for long periods.|
