|Drug Name and Strength||Enrofloxacin 150mg|
|Indication||Treatment of bacterial infections caused by organisms sensitive to enrofloxacin in dogs, including infections of the skin, upper and lower respiratory tract, genitourinary tract, wounds, abscesses and discharging sinuses.|
|Contraindications||Administration to dogs during the rapid growth phase. This means enrofloxacin should not be used in dogs under 1 year of age, or under 18 months of age for giant breeds. |Actions||Enrofloxacin is a synthetic drug from the class of fluoroquinolones. It has a broad spectrum of bactericidal activity against Gram-negative and Gram-positive bacteria, including Staphylococcus aureus, Staph. intermedius, Streptococcus sp., Actinomyces pyogenes, Escherichia coli, Klebsiella sp., Proteus sp., Pseudomonas aeruginosa, Brucella canis, Salmonella sp., Bordetella bronchiseptica and Mycoplasma sp. Enrofloxacin exerts its effect on bacterial DNA-gyrase. This is a type II topoisomerase enzyme that plays a crucial role in the unwinding, cutting and resealing of DNA. Inhibition of this enzyme leads to replication failure.Enrofloxacin is rapidly absorbed from the gastrointestinal tract and has a high volume of distribution, reaching high concentrations in the liver, bile, kidney, lungs and reproductive system including prostatic tissue and fluid. Therapeutic levels are also attained in bone, synovial fluid, skin, muscles, aqueous humour and pleural fluid. Low concentrations only are reached in the cerebrospinal fluid. Peak serum levels of enrofloxacin are reached within one hour of oral dosing.Elimination of enrofloxacin is via a combination of renal and nonrenal routes. Fifteen to 50% is excreted unchanged in the urine following tubular secretion and glomerular filtration. The remainder undergoes metabolism to various metabolites that are less active than the parent compounds. Thirty to 40% of the circulating enrofloxacin is metabolised to ciprofloxacin. Metabolites are excreted in urine and faeces.The presence of food may delay the rate of absorption; however, the extent of absorption is not affected. Drug performance is both concentration and area-under-the-curve dependent. Enrofloxacin is considered to be Category B for pregnancy: limited information available including some evidence of effects on fetus in laboratory animals from some drugs in this class. Enrofloxacin passes into breast milk.|
|Precautions||Enrofloxacin can be used in patients with liver disease, with careful monitoring. Use with caution in animals with pre-existing kidney disease.First Aid If poisoning occurs, contact a doctor or Poisons Information Centre. Phone Australia 131 126.|
