Indication | Treatment of canine congestive heart failure (CHF) originating from dilated cardiomyopathy (DCM) or valvular insufficiency (mitral and/or tricuspid regurgitation). Treatment of preclinical DCM in large breed dogs, to prolong the time to the onset of CHF or death. Treatment of dogs with cardiomegaly secondary to preclinical myxomatous mitral valve disease, to reduce heart size and increase survival time. |
Contraindications | This product is contraindicated in cases of hypertrophic cardiomyopathy or clinical conditions where an augmentation of cardiac output is not recommended for functional or anatomical reasons (e.g. aortic stenosis). |
Actions | Pharmacology Pimobendan exerts its stimulatory myocardial effect by a dual mechanism of action: increase in calcium sensitivity of cardiac myofilaments and inhibition of phosphodiesterase (type III). It also exhibits a vasodilatory effect through an inhibitory action on phosphodiesterase III activity.Pharmacokinetics Following oral administration of pimobendan, the absolute bioavailability of the active principle is 60 to 63%. The mean plasma protein binding is 93%. The plasma elimination half-life of pimobendan is approximately 30 minutes, and the main active metabolite elimination half-life is approximately two hours. Almost the entire dose is eliminated via faeces. |
Precautions | Use during pregnancy and lactation. In studies with rats and rabbits pimobendan had no effect on fertility, and embryotoxic effects only occurred at maternotoxic doses. In rat experiments it has been shown that pimobendan is excreted into milk. Vetmedin Chewable Tablets should only be administered to pregnant and lactating bitches if the expected therapeutic benefit outweighs the potential risk.First Aid If poisoning occurs, contact a doctor or Poisons Information Centre. Dosage and Administration | Administer orally at a dose range of pimobendan 0.1 to 0.3 mg/kg twice daily with an ideal dose of 0.25 mg/kg twice daily. Each dose should be given approximately one hour before feeding. 2 to 4 kg bodyweight. Half a 1.25 mg tablet (morning); half a 1.25 mg tablet (evening). 5 to 10 kg. One 1.25 mg tablet (morning); one 1.25 mg tablet (evening). 11 to 20 kg. Half a 5 mg tablet (morning); half a 5 mg tablet (evening). 21 to 40 kg. One 5 mg or ½ 10 mg tablet (morning); one 5 mg or ½ 10 mg tablet (evening). 41 to 60 kg. Two 5 mg tablets or one 10 mg tablet (morning); two 5 mg tablets or one 10 mg tablet (evening). 60 to 80 kg. Three 5 mg or one and a ½ 10 mg tablets (morning); three 5 mg or one and a ½ 10 mg tablets (evening). > 80 kg. Two 10 mg tablets (morning); two 10 mg tablets (evening).Vetmedin chewable tablets may be combined with a diuretic treatment such as furosemide. |
